A June 15 systematic review in the American Journal of Clinical Dermatology pooled 19 studies and 67,568 patients and linked GLP-1 receptor agonists to real improvement in hidradenitis suppurativa, a painful inflammatory skin disease. Sixty percent of patients dropped at least one Hurley severity stage, quality-of-life scores improved by 3.83 points, and C-reactive protein and HbA1c both fell. The catch that makes it interesting: average BMI fell only 2.64 kg/m2, too little to explain the skin gains on its own, hinting the drugs' anti-inflammatory side is doing real work, though the mostly observational evidence cannot prove it.

A meta-analysis of 59 real-world studies and 24,859 patients found the pill form of semaglutide lowered blood sugar and weight by about as much as the registration trials promised. The catch: the studies disagreed enormously, so the direction is solid but the exact size is soft.

This week's headline fix for muscle loss on obesity drugs was an antibody, apitegromab, in Nature Medicine. A day later a University of Alberta mouse study showed an oral ketone ester preserved muscle on semaglutide without blunting fat loss. The cheapest answer yet, and still only preclinical.

Innovent's mazdutide beat semaglutide in a head-to-head diabetes trial, with more than twice as many patients hitting both their blood-sugar and weight goals. Its obesity trial, published in JAMA, showed 16.65 percent weight loss.

The 16.6 percent topline was old news. The full SYNCHRONIZE-1 data, out at the diabetes meeting and in the New England Journal of Medicine, showed visceral fat down 34 percent, liver fat down 63 percent, and lean mass largely spared. The glucagon arm is why.

Full Phase 3 REIMAGINE 2 data at ADA 2026 show Novo's amylin-plus-GLP-1 combination cut weight 14.2 percent versus 10.2 percent for semaglutide by itself, while the blood-sugar improvement barely moved. The split shows exactly what the amylin half buys.

Structure Therapeutics published full Phase 2b data for aleniglipron, a once-daily oral small molecule, in Nature Medicine. On the top dose, 70 percent of people lost at least 10 percent of their body weight, and the loss kept deepening past 36 weeks.

A post hoc analysis of SURMOUNT-1 went back to stored blood from 392 participants and measured 17 cardiovascular markers. At 72 weeks the top dose of tirzepatide cut inflammation (hsCRP -54.6 percent), leptin (-61.4 percent), and clotting protein PAI-1 (about -40 percent), and raised protective adiponectin (+47.7 percent), with most effects deepening step by step from 5 to 10 to 15 mg.

In obese mice, semaglutide stripped skeletal muscle as it stripped fat and left newly healed fibers undersized. Adding a 15-PGDH inhibitor woke muscle stem cells and recovered strength without slowing the weight loss.

AstraZeneca's eleglipron, the oral small-molecule GLP-1 agonist licensed from Eccogene in 2023, met primary endpoints in Phase 2b VISTA (310 obesity patients, 26-week weight loss) and SOLSTICE (406 T2D patients, 26-week HbA1c vs semaglutide). Full data at ADA 2026 in June. AstraZeneca commits to a comprehensive Phase 3 program, adding a third serious entrant to the oral GLP-1 race.

Lilly disclosed an ADA 2026 abstract slot for animal data on a unimolecular quintuple agonist engaging GLP-1, GIP, glucagon, amylin, and calcitonin receptors. The compound is distinct from the DiMarchi-Tschöp-Müller GLP-1R/GIPR/PPAR conjugate covered May 1, but the convergence of two independent groups producing five-pharmacophore obesity drugs in the same publication window is a real signal about where multi-agonist development is heading.

Novo's high-dose Wegovy (semaglutide 7.2 mg injection), FDA-approved in March and launched April 7, contributed to the Q1 print on STEP UP trial data showing 21% mean weight loss at 72 weeks and 89% of patients achieving at least 5% body-weight reduction. Dysesthesia at 22% (vs 6% on 2.4 mg and 0.3% placebo) is the safety chapter prescribers will need to engage explicitly. Step-up requires four-plus weeks tolerability on standard dose first.

Eli Lilly disclosed positive topline results from TRANSCEND T2D1 on the April 30 Q1 call. The triple GLP-1/GIP/glucagon agonist lowered A1c by 1.7 to 2.0 percentage points and produced mean weight loss of 25 to 37 pounds versus placebo in adults with type 2 diabetes. Seven more Phase 3 readouts are expected in 2026, with regulatory submission targeted for late 2026 or 2027.

Boehringer and Zealand reported 16.6% mean weight loss at 76 weeks for survodutide in Phase 3 SYNCHRONIZE-1, versus 3.2% on placebo, with predominantly fat-mass loss and 85.1% of treated patients clearing the 5% threshold. The same announcement advanced BI 3034701, a triple GLP-1/GIP/NPY2 agonist, into Phase 2 mid-2026.

Novo Nordisk's PIONEER TEENS Phase 3a trial in 132 kids aged 10 to 17 with type 2 diabetes cut HbA1c by 0.83 percentage points more than placebo at 26 weeks. Filing for label expansion is planned for H2 2026. It would be the first oral GLP-1 receptor agonist in this age group.
