Two-thirds of the published research behind six of the most-sold "recovery" peptides comes from animals, mostly rats. The human trials that exist are few and small, and most ran without proper controls. That is the finding of a scoping review published August 11 in the American Journal of Sports Medicine ↗. The journal is one of the field's higher-bar titles, and the review lands on the exact compounds that fill gray-market shopping carts.
The reviewers worked through the PubMed database under PRISMA screening rules, the standard checklist for systematic evidence reviews. They pulled every study they could find on six peptides marketed for musculoskeletal recovery and performance. The list: BPC-157 ↗, TB-500 ↗ (a synthetic version of the protein thymosin beta-4), CJC-1295 ↗, MK-677 (ibutamoren), ipamorelin ↗, and GHK-Cu, the copper-binding peptide ↗. These are the names traded in fitness forums and sold by compounding and "research chemical" vendors. The pitch is usually that they speed healing of tendon, muscle, bone, and ligament, and that they carry little risk.
Promising in rats, thin in people
In animal models the picture was encouraging but uneven. Each compound showed its own mechanism and some effect on healing tissue, which is how these peptides got their reputations in the first place. The problem is what happens when the review looks for the same effects in humans. There, the studies thin out fast. The authors describe the human data as heterogeneous and, at best, showing modest improvement for two narrow uses: metabolic bone health and degenerative knee pain. Everything else marketed as proven is, by this review's reading, not.
That gap between the marketing and the evidence is the story. A peptide that helps a rat's Achilles tendon heal after a surgeon cuts and repairs it is a real result. It is not the same claim as a vial that will fix a weekend athlete's tendinopathy. The dosing and the delivery route also swing wildly across studies. Even the animal wins are hard to translate into a number a person could act on.
One of the six carries a real risk
The review does not treat all six as merely unproven. It singles out MK-677, an orally active drug that ramps up growth-hormone release, for documented harm. The named risks are an association with congestive heart failure and broader metabolic dysfunction, including insulin resistance. Worth a note for accuracy, MK-677 is not actually a peptide. It is a small-molecule ghrelin-receptor agonist that gets sold alongside the peptides and lumped in with them. The point stands either way. The one compound in the set with the clearest human signal points toward risk, not benefit.
The authors' conclusion is blunt for a review. Because the efficacy and safety data are not there, they write, these supplements should not be recommended as a replacement or an add-on to standard orthopedic care.
Why this belongs on a peptide site
A scoping review maps the evidence rather than pooling it into a single effect size, so it cannot tell you how well BPC-157 works, only how little has actually been tested. That is the honest limit here. But mapping is exactly what the gray-market peptide space has lacked. Buyers see testimonials and mechanism diagrams, not a count of how many controlled human trials exist, which for most of these is close to zero.
